This EIA kit is the third generation double antigen sandwich method for the detection of circulating antibodies to Human Immunodeficiency Virus Type 1 (HIV-1) and/or Human Immunodeficiency Virus Type 2 (HIV-2) in Human serum or plasma and is indicated as a screening test for serum or plasma and as an aid in the diagnosis of potential infection with HIV-1 and/or HIV-2.

Summary and Principle of the Test

The Human Immunodeficiency Virus type 1 (HIV-1) has been isolated from patients with acquired immune deficiency syndrome (AIDS) and AIDS-related complex (ARC). HIV-1 has been considered the sole pathogen of these syndromes until another human immunodeficiency virus (HIV-2) was isolated in 1986, reportedly also capable of causing AIDS. Since its initial discovery, over 600 cases of HIV-2 infection have been documented worldwide, with over 40 AIDS cases associated with HIV-2.

Both viruses share similar morphology and lymphotropic properties, and their modes of transmission appear to be similar. Furthermore, HIV-1 and HIV-2 genomes exhibit approximately 60% homology in conserved genes such as gag and pol. Serological studies also indicate frequent cross-reactivity between core proteins of HIV-1 and HIV-2, while envelope proteins are more type-specific.

Despite this immune cross-reactivity, there are significant differences in detecting HIV-2 antibodies using any licensed HIV-1 enzyme immunoassay (EIA). The HIV-1/HIV-2 EIA is designed to detect HIV-1 and/or HIV-2 antibodies for blood screening and diagnostic purposes.

Any sample that reacts with the HIV-1/HIV-2 EIA in initial testing must be retested using another company's HIV-1/HIV-2 EIA. Samples with repeat reactive results may contain HIV-1 or HIV-2 antibodies. Therefore, additional, more specific, or supplementary tests for HIV-1 and HIV-2 antibodies, such as immunoblotting, immunofluorescence, or radioimmunoprecipitation, must be conducted to confirm the presence of HIV antibodies.
